Originally researched and developed at the University of Arizona as a possible sunless tanning solution to help prevent skin cancer by causing production of melanin without UV exposure, Melanotan II is a synthetic analog of the natural melanocortin peptide hormone alpha-melanocyte stimulating hormone (α-MSH), which was found to have too short a half-life to be effective as a therapeutic drug. Melanotan II was isolated and upon evaluation was determined to be 1,000 times more potent than its naturally-occurring counterpart. A cyclic lactam analog of α-MSH, it has the amino acid sequence Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2. Melanotan II has been used in a variety of medical studies and clinical trials related to the primary reason for its isolation, to provide melanogenesis effects. It has also been used for a primary treatment in a variety of medical studies and clinical trials, including topics such as vagal outflow to the stomach, its aphrodisiac qualities, changes in melanocytic lesions on a patient afflicted with FAMMM syndrome, the role Melanotan II may play in priapism cases and food intake reductions.
